Gene Therapy Vaccines Biotherapeutics Proteins Nanoparticles Polymers SEC-MALS Measure size, titer (capsids/mL), aggregated fraction, and relative capsid content (empty:full ratio) of viral vectors with a DAWN ™ MALS instrument, an Optilab ™ refractive index detector and a HPLC UV module. Field-Flow Fractionation Resolve and analyze viral vectors, LNP-RNA particles and large AAV aggregates using the Eclipse ™ field-flow fractionation system combined with online MALS and DLS. DLS For size, size distributions and titer (capsids/mL), use the DynaPro ™ NanoStar ™ or DynaPro ™ ZetaStar ™ cuvette-based instrument to test a few samples, or a DynaPro ™ Plate Reader when many samples, conditions and replicates are required. AAV Method Implementation & Training To get up and running onsite with a turnkey SEC method and standard operating procedure, the AAV Method Implementation & Training service is key for characterizing key gene therapy attributes. Viruses and VLPs Screen formulations and processes for stability, aggregation and physical titer with HT-DLS. Gene Vectors Quantify DNA/RNA content, particle concentration and size distributions of viral and non-viral vectors with SEC/FFF-MALS. Subunit Antigens Characterize oligomeric state, antibody binding affinity and absolute stoichiometry by SEC-MALS and CG-MALS. Polysaccharides Develop, monitor and control production processes more effectively with real-time MALS. Aggregation While SEC-MALS with a DAWN ™ or miniDAWN ™ is the primary means of characterizing soluble protein aggregates, FFF-MALS, CG-MALS and DLS are valuable orthogonal techniques to meet FDA requirements. Conjugation SEC or FFF plus triple detection UV-MALS-RI and ASTRA ™ software determine the molar mass of protein and modifier in conjugated biotherapeutics such as antibody-drug conjugates or PEGylated proteins. Formulation The DynaPro ™ Plate Reader performs high-throughput screening of excipients and buffers for optimal formulation in situ in standard microwell plates, detecting aggregation as well as conformational and colloidal stability indicators. Interaction CG-MALS combines a Calypso ™ and DAWN or miniDAWN to characterize the affinity and absolute molecular stoichiometry of drug-target binding, without labeling or immobilization. It also analyzes self-association at low, moderate and high concentrations. Identification A DAWN ™ or miniDAWN ™ MALS detector, combined with a Wyatt SEC column and the Optilab ™ dRI detector, determine absolute molar masses and sizes of peptides, proteins and other biomacromolecules independently of column calibration. Purification Validate protein purification and stability quickly and easily with a DynaPro ™ NanoStar ™ or DynaPro ™ ZetaStar ™ cuvette-based, combined dynamic light scattering & static light scattering detector, or a high-throughput DynaPro ™ Plate Reader. Charge The ZetaStar ™ is the only electrophoretic light scattering detector capable of providing reliable and repeatable measurements of a protein’s net charge in high-salt buffers. Binding CG-MALS combines a Calypso ™ and DAWN or miniDAWN to characterize the affinity and absolute molecular stoichiometry of simple or complex biomolecular interactions. CG-MALS requires no labeling or immobilization, addressing self-association as well as hetero-association. Accurate Size Distributions Accurate size distributions with true ensemble representation require fractionation + independent, on-line determination of size. A DAWN ™ / Eclipse ™ FFF-MALS system can characterize simple or complex fluids incorporating macromolecules with large and small nanoparticles. Conformation and Composition Information about nanoparticle shape and structure can be obtained by combining MALS analysis with dynamic light scattering. The WyattQELS ™ DLS module integrates into the DAWN ™ or miniDAWN ™ for simultaneous MALS and DLS analysis. Add a UV, fluorescence and/or Optilab ™ dRI detector for compositional studies. High-Throughput Sizing Need to characterize your nanoparticles under hundreds of solvent conditions in the space of a few hours? The DynaPro ™ Plate Reader measures size distributions by DLS, in situ in standard 96-, 384- or 1536-well microtiter plates. A camera conveniently photographs each well to identify precipitates. Zeta Potential / Charge The ZetaStar ™ is the only zeta potential detector capable of hands-free, high-throughput measurements by coupling to an autosampler. It measures charge and size simultaneously to monitor aggregation or degradation caused by the applied electric field. Absolute Molar Mass and Size A DAWN ™ MALS detector and Optilab ™ dRI detector work together to determine distributions of absolute molar mass and size of natural or synthetic polymers independently of column calibration . Branching and Conformation Incorporate a WyattQELS ™ DLS module and/or a ViscoStar ™ differential viscometer to characterize the branching ratio and conformation of complex polymers. Biodegradation Light scattering is an excellent means for assessing the degradation of biopolymers as a result of exposure to heat, light, high or low pH, and other stimuli. dn/dc and Zimm plots Wyatt offers several options to simplify laborious, error-prone dn/dc and Zimm analyses for natural and synthetic polymers. What can I measure? Molar Mass Size Charge Interactions Conformation Conjugation Wyatt Technology Wyatt Technology is the recognized leader in light scattering instrumentation and field-flow fractionation systems for determining the absolute molar mass, size, charge and interactions of macromolecules and nanoparticles in solution. Wyatt’s scientific tools support the pharmaceutical industry, academia and government labs worldwide, in the research and development of novel therapeutics including vaccines, gene vectors, nano-drug carriers, monoclonal antibodies, and other advanced pharmaceuticals, as well as fundamental life and material sciences. be_ixf;ym_202405 d_14; ct_100 be_ixf; php_sdk; php_sdk_1.4.26 https://www.wyatt.com/ https://www.wyatt.com/ Ultimate Guide Thirteen key questions to ask when buying a Multi-Angle Light Scattering (MALS) or Dynamic Light Scattering (DLS) instrument.
